Introduction
Pluripotent stem cells (PSCs) have the unique capability to proliferate indefinitely and to differentiate into all cell types. iPSCs were found to be very similar to ESCs, which ESC-like colony morphology and expression of pluripotency markers. During the reprogramming process, the transcriptional network changes gradually from the somatic to the pluripotent state. The fully reprogrammed cells showing the most ES cell like patterns of gene expression, and partially reprogrammed cells showing intermediate phenotypes. The classical markers for the pluripotent embryonic state, such as alkaline phosphatase activity or TRA expression, are critical for tracking cell reprogramming and for gaining confidence in the dedifferentiated status of the reprogrammed cells.
