An Overview of WD repeat-containing protein 5 (WDR5)
WD repeat-containing protein 5 (WDR5) is a member of the WD-40 protein family and is also an important component of the chromosomal H3 modified protein complex Trithorax group (Trx G). WDR5 is composed of 333 amino acids, and its peptide chain contains 7 WD repeats to form a β-sheet, which is the main functional domain of WDR5. As a member of the WD40 family, it is widely involved in the regulation of gene expression in organisms, and plays a huge role in the activation of mixed lineage leukemia, tumor formation, assembly of Mitochondrial antiviral-signaling protein (MAVS) -related signaling complexes, and activation of Interferon regulatory factor 3 (IRF3) and nuclear factor-kappa B (NF-κB). WDR5 plays an important role in the maintenance, differentiation, and tumorigenesis and development of embryonic stem cells. Down-regulation of WDR5 can inhibit the proliferation of osteoblasts, while up-regulation of WDR5 can promote the proliferation and self-renewal of bladder cancer cells.
