An Overview of Vesicular monoamine transporter 2 (VMAT2)
Vesicular monoamine transporter 2 (VMAT2) is a protein that transports neurotransmitters and belongs to a subtype of vesicular monoamine transporters. VMAT2 is a glycoprotein that binds to the vesicle membrane and is an essential protein that constitutes the human body. It transports monoamines, especially dopamine, norepinephrine, serotonin and histamine, from the cellular cytoplasm to synaptic vesicles. VMAT2 has approximately 518 amino acids. The Vesicular monoamine transporter’s relative molecular mass and the number of amino acids in different species are slightly different. Prediction of its molecular structure from the VMAT2 protein sequence revealed that VMAT2 has 12 transmembrane helix regions with N2 terminus and C2 terminus in the cytoplasm. VMAT2 is expressed in dopaminergic, noradrenergic, adrenergic, serotonergic, serotonergic, and histaminergic neurons and sympathetic nerves in the human brain. In addition, VMAT2 is expressed in mast cells, megakaryocytes, platelets, basophils, and Langerhans cells in the hematopoietic system. VMAT2 is a proton-dependent transporter that relies on a transmembrane H+ electrochemical gradient to provide energy to mediate the transport of monoamines in vesicles, including norepinephrine, dopamine, and serotonin.
