Introduction
CRISPR/Cas9 technology has been used as a genome-editing tool for multiple cell lines, such as tumor cell line and embryonic stem cell lines. Primary T cell, as an important class of immune cells system, play an extremely important role in immunity system. It holds great potential for therapeutic applications to engineer genome in primary T cells. However, genome engineering in primary T cells is still a challenge due to low delivery efficiency and limited expression of CRISPR/Cas9 components.
